Ackles really stands out as the Red Hood. The Supernatural star makes the character arrogant, funny, and tragic...the combination is a perfect match for Jason Todd. Source

Sera Gamble gives interviews and reveals some of the direction of Season 6 with an empahsis on family origins and the fact that it will have a noir feel to it:

I'm a huge noir fan, Bob Singer are huge noir fans. Eric loves this stuff too. It lends a different kind of gritty darkness because the heroes in those stories are not pure white hats.
If you think of 'L.A. Confidential,' if you think about Sam and Dean together being like a Bud White or being like a Bud White…. Bud White beats people up. He has anger management problems. He drinks too much. But he's a hero. The fact that he is moral is a problem. The other sort of hero in that story has a sort of moral relativism.

"There are a lot of shades of gray that we're playing with this season, in terms of the kind of heroes we're interested in."

She was asked whether stepping into the showrunner role meant Sam and Dean would be getting their gear off more:

"I think the shirtlessness ratio is the same," Gamble laughs. "It's cold in Vancouver! I have to put their health first." Source

Leading up to the Season 6 premiere, Jared and Jensen give an interview to KSiteTV about what to expect and how their characters have changed.

JARED: I think it kind of hearkens back to Season One where there’s Sam’s story, and Sam’s like “Dean, with all due respect, I deserve respect as well. I have my own life. I’ve done my own things. I deserve my own credit, and I’m going to do things my way, and I deserve to do things my way.” And Dean’s like, “well, you don’t know what I’ve been through in this last year, and my priorities have changed and shifted. Now I don’t want to be so cavalier.” And Sam’s like “What? Why not? I just came back from Hell again!” So I think there are personal frictions with both brothers, and they’re sort of amplified by the fact that we hunt.

JENSEN: There was a big kind of a shift in Dean’s character, actually much to my chagrin. I was not happy with it. Dean was really kind of being written soft. We spent five years with this guy being a tough, shoot-first-ask-questions-later kind of guy, and now all of a sudden he’s lived one year in more of a domesticated life, and he’s gone soft on us. As a fan of the show myself, that kind of made me upset, but I did my best to kind of curve that in the acting. It read pretty soft on the page, but I think I toughened it. I’m like “Look, if I’ve got anything to do with it, I’m gonna beat this guy up a little bit.”

JARED: In all honesty, you kind of kept it true to the character. It wasn’t like you were just not doing what the writers were writing for. It was like “How can I work this in to where I don’t completely lose….?”

JENSEN: Which was different, because after five seasons of playing this guy, now I actually have to think harder about how to play this guy correctly than I have before.

November 2 Polling the Electorate!
Tv Guide runs a competition where fans can vote for their favourite show to be on the TV Guide Cover. In the final days the race was between Supernatural and Chuck - which scores an average nearly 3 times as many viewers. Supernatural fans cried foul when it was discovered that Chuck fans were using a macro that automatically reloaded the page and repositioned the cursor for faster voting.

During the final hours of voting Showrunner Sera Gamble thanked fans:

Hey-- just a quick note on behalf of Bob, Eric, myself, and the whole gang here at Supernatural. We want to thank our incredible fans for your support. We hear you've been keeping us in the running against some very worthy competition. Regardless of what happens, we're deeply touched as always that you've got our backs. (But yeah, of course we'd love to see Jensen and Jared on the cover -- and we think they couldn't deserve it more -- so please do keep voting for 'em.) Thanks again, guys!

Source.

Supernatural won the competition and were featured on the December 16th cover of TV Guide. The cast thanked the fans in a video.
