The Ark of the Covenant is thought to be one of the weapons or "nukes" of Heaven. It is a golden chest said to contain, amongst other Biblical relics, the original stone tablets containing the Ten Commandments that Moses received from God. Though it has not appeared on the show, it has been implied that it too is missing from Heaven, alongside the Staff of Moses, Lot's Salt, and Gabriel's Horn of Truth. Sam lures Castiel to him by falsely claiming that they have found a lead on the Ark's location (using the premise of the blockbuster Raiders of the Lost Ark as a guide).6.10 Caged Heat

Ark of the Covenant in Lore
The Ark of the Covenant was crafted following the Exodus of the Hebrews out of Egypt under the leadership of Moses. Its primary purpose was to hold, among other Biblical artifacts, the stone tablets containing the Ten Commandments that Moses brought down from Mount Sinai. It was described as a golden chest measuring 2.5 cubits in length, 1.5 cubits in breadth, and 1.5 cubits in height. It is said to have been plated entirely with gold, with a crown or molding of gold to be put around it. Four rings of gold are said to be put into its four feet, with two on each side, and through these rings staves of shittim-wood overlaid with gold for carrying the Ark are to be inserted, and these are not to be removed. A golden cover, adorned with two golden cherubim, is said to be placed above the Ark.
In one story of the Ark, it is said that the Israelites conquered the city of Jericho by marching around the city's high walls carrying the Ark before them for seven days by seven priests sounding seven trumpets of rams' horns. At the end of these seven days, the walls of the city collapsed, allowing the Israelites to take the city with ease.
