The term "Apocalypse" is commonly and most often used to describe events and disasters that will occur during the end of the world.

Apocalypse in Canon

In Supernatural, the Apocalypse or Armageddon is started when The 66 Seals that bind Lucifer are broken and he is freed. Zachariah states that when the apocalypse starts Four Horsemen will arise, red oceans and fiery skies will be witnessed5.01 Sympathy For The Devil

The First Seal will break when a righteous Man sheds blood in Hell; Dean breaks this seal when he spills the blood of a human soul in Hell.
The Last Seal will break when Lucifer's first demon is destroyed; Sam breaks this seal when he kills Lilith.

Many members of the Host of Heaven like Zachariah and Michael wanted The 66 Seals broken so that Lucifer can break free from his Cage, which would give Michael the opportunity to kill him. Raphael states that he wants paradise and he also supports the his older brother's plan to release Lucifer so that Michael can destroy him. Michael believes that he can establish paradise by killing Lucifer. Michael sees killing Lucifer as his destiny, although he professes great love towards Lucifer.

Castiel and Zachariah tell Dean that he will stop Lucifer. Zachariah later revels that Dean alone cannot possibly stop Lucifer, but he tells Dean that he is Michael's true Vessel.

The Four Horsemen of the Apocalypse, the Croatoan Virus and the Whore Of Babylon all served as instruments of destruction during the Apocalypse. Jesse Turner, the Anti-Christ, according to the Book of Revelations, will also play an important role during the destruction of Earth, but he decided to turn away from Lucifer's plans.

Following soon after the events of5.22 Swan Song, a cival conflict in Heaven amongst angels brews. Currently, Raphael is tying to manage Heaven due to Michael's absence. He ordered a group of angels to aid him in searching for and gathering God's weapons, in which case would somehow release Lucifer and Michel, effectively restarting the Apocalypse but by unspecified means6.03 The Third Man
