As the Darkness barreled towards and engulfed the Impala, Dean sees the flash of a woman in a black dress in front of him before he disappears, and Sam is knocked unconscious.
Dean questions this manifestation of the Darkness - Amara. She is surprised Dean knows about the Darkness, and she claims to know nothing of Death. Amara tells him that they are bound and will always help each other, revealing that she bears the Mark of Cain.
Sam, Dean and Deputy Jenna Nickerson also rescue a newborn child, from Mike Schneider who is infected by one of the rabids. Just before he dies, he says that the child is called Amara. She also bears a tiny birthmark in the shape of the Mark of Cain.
