Dean introduces himself to Becky as Sam's older brother.

Dean tells Michael he can related to him because he also is a older brother.

Dean: Hey Michael. Hey. I know how you feel - I'm a big brother too - but you gotta go easy on your Mom right now, ok?

Michael: You said you were a big brother.
Dean: Yeah.
Michael: You'd take care of your little brother? You'd do anything for him?
Dean: Yeah I would.

Sam tells Dean he'll do whatever it takes to get him out of his deal.

Sam tells Dean he still believes in him because he's his big brother.

Dean: I—I don’t get it. Sam, why are you doing this?
Sam: Because… You’re still my big brother.
